Guidelines for HIR Central Facility: AKTA Purifier Plus 10
The AKTA Purifier Plus 10 versatile, modular liquid chromatography systems for fast and reliable
separations of proteins and peptides.
The Committee reserves the right to make any necessary changes to the guidelines from time to time and as
and when needed.
Location
The AKTA Purifier Plus 10 is at present located at the HIR building, central facility 2.
Operating Hours
The AKTA Purifier Plus 10 is available for service from 10 am to 5 pm, working weekdays. Advance
appointments are required. Time could be booked up to two weeks in advance.
Online Booking
Only qualified users will have access to the booking calendar. You must book online to utilize the machine.
Please plan your experiments ahead and make sure the facility can accommodate your schedule. The slots are
scheduled on a first-booked-first-served basis. Confirmation is done at the discretion of the officer in charge,
to maximize availability for the entire UM community. Please keep your appointments, and show up for them
promptly. User has to complete the experiments within the time slot allocated. Failing to do so, the officer has
the right to abort the experiment and vacate the machine for the next user. Cancellation of booking must be
made at least two working days before the actual equipment operation time. You will be billed for 50% of the
charges for late cancellations. Users will have to fill up a letter of intent available online.
Export of Data
If necessary, data can be saved using CD (supplied by the user) only.
Charges
The AKTA Purifier Plus 10 is FOC for UM-researcher and RM20 per run for non-UM researchers if
researcher bring their own columns. Columns are available for rent :

Guidelines for using the HIR Atomic Emission Spectrometry (AES)
The HIR Agilent 4100 MP-AES runs unattended without flammable or expensive gas supply. This unit
runs entirely on air, which means one can perform automated multi-element analyses while eliminating
flammable and expensive gases. This unit offers:

Powerful performance - The magnetically excited microwave plasma source provides detection
limits down to sub ppb levels.

Better reliability - The robust system is built for a range of industries including Toxicology
studies, Geochemistry, Food Testing and Agriculture, Energy and Fuels, and Environmental, and is
ideal for remote environments.

Guidelines for using the HIR Bioanalyzer Expert 2100
The HIR Bioanalyzer Expert 2100 is a multi-user facility which provides fast and user friendly
instrumentation to researchers for analyzing nucleotide and protein’s concentration and purity.
All consumable (Reagent and Chip) shall be provide by user. Before and after usage of the
Bioanalyzer, user are responsible to clean the detector head with clean ultrapure water with the
particular chip.

The HIR Bioanalyzer is located at Central Facility 2, Level 1, HIR Building

The HIR BIOLOG Facility is a multi-user facility which provides advanced, comprehensive
automated high throughput phenotype microarray system for quantitative phenotypic cellular
assays with microbial cells and mammalian cells; integrated for automatic characterization and
identification technology of bacteria [aerobic and anaerobic], yeasts and filamentous fungi. The
Committee reserves the right to make any necessary changes to the guidelines from time to time
and as and when needed.
Location
The HIR BIOLOG Facility is at present located at the 1st Floor Centre HIR lab, HIR Building.
Operating Hours
The HIR BIOLOG Facility is available for service from 10 am to 5 pm, working weekdays.
Advance appointments are required. Time could be booked up to two weeks in advance. Strictly no
block booking is allowed. Slots for usage are set at three-hours each i.e. 10 am – 1 pm and 2 pm – 5
pm, or from 10 am - 5pm. The number of people that can be present from each user laboratory is
limited to two, except by specific pre-arrangement with the facility staff.

The HIR Biomark HD and C1 Single-Cell auto Prep System facility is a multi-user facility which provides resarchers
with cutting edge Biomark HD and C1 Single-Cell auto Prep System facility for latest research in single-cell study.
The Committee reserves the right to make any necessary changes to the guidelines from time to time and as and
when needed.
Location
The HIR Biomark HD and C1 Single-Cell auto Prep System facility is at present located at Central Facility 2.2,
Level 2, HIR Building.
Operating Hours
The HIR Biomark HD and C1 Single-Cell auto Prep System facility is available for use from 9 am to 5 pm, working
weekdays. Weekend or holdiay booking requires a separate booking that user will have to inform by email the lab
officer and responsible officers. Advance appointment is required. Time could be booked up to 1 day in advance.
Walk-in usage will only be allowed provided there is no prior booking and in such instance, the user will still need
to do the booking. No block booking is allowed.
Online Booking
Only qualified users will have access to the booking calendar. You must book online to utilize the machine. Please
plan your experiments ahead and make sure the facility can accommodate your schedule. The slots are scheduled
on a first-booked-first-served basis. Confirmation is done at the discretion of the officer in charge, to maximize

The HIR Cary 60 UV-VIS facility is a multi-user facility which can handle the widest variety of
sample sizes and types with liquid sampling accessories including fiber optic probes and
couplers, Peltier and water thermostatted single and multicell holders, temperature probes,
microvolume sampling cells and rapid mix accessory for stopped flow kinetics. For solid
samples, choose from solid sample holder, fiber optic reflectance probe and coupler and fixed
angle specular reflectance accessories (SRA).

The HIR Cary 60 UV-VIS facility is at present located at Central Facility 1, Level 1, HIR
Building.

Identification System
The Agilent Gas Chromatography-MIDI Microbial Identification System is a multi-user facility
that offers researchers with identification of bacteria based on analysis of cellular lipidic fraction
of bacteria such as Fatty Acid Methyl Esters (FAME). Determination of the FAMEs was
coupled with the use of Agilent Gas Chromatography hence it allows rapid bacterial
identification without the requirement of biochemical kits. All consumables and sample
preparations have to be provided by user. Users are responsible to make sure the machine is not
contaminated with their sample (especially bacteria extracts) after usage. User are adviced to
switch on the instrument 30 mins before use.

Guidelines for Using the HIR Agilent 5975C GC/MSD Facility
Gas chromatography-mass spectrometry (GC-MS) is an instrument that separates chemical compounds in a mixture (GC
component) and identifies the separated compounds by their mass over charge ratio (MS component). GC-MS is used in a
wide range of applications including analysis of lipids and drug metabolites, environmental analysis and identification of
chemical compounds in a mixture. One of the advantages of GC-MS is its ablity to provide identification of chemical
compounds based on the mass spectra provided in the incorporated library.

Sample Preparation Requirements
 The samples MUST BE ABLE TO VOLATILISE at temperatures below 300C. Thus, samples of total plant extracts,
untreated waste-water, crude oil, strong acid or base solutions, salts, metals and polymeric materials are not suitable to
be analysed by GC-MS.
 Samples must be prepared in guranteed grade (GR) or GC-MS grade solvents, preferably acetone, methanol or hexane.
Less preferable are halogenated solvents (e.g., chloroform, methylene chloride, etc.) and ethers (e.g., diethylether,
THF). DO NOT use water, DMF, or DMSO as solvent.
 The upper limit of sample concentration is 100.0 ppm in 0.5–1.5 mL. Overloading samples may cause damage to the
ion source in the MS component.
 Every sample must be filtered through a 0.45 µm membrane filter before injection into the GC-MS.
 Every sample must be placed in a GC-MS specified sample vial with the correct septum and cap, i.e., Agilent Part
Number 5182-0714 (Pack of 100 screw top vials) and Agilent Part Number 5182-0717 (Pack of 100 blue screw caps
and PTFE/red rubber septas).

The HIR ITC allow characterization of biomolecular interactions by providing direct, label-free
measurements of binding affinity and thermodynamics. ITC provides valuable information about
binding interactions, molecular stability, protein folding, and enzyme kinetics. ITC measures all binding
parameters in a single label-free, in-solution experiment. Parameters include binding affinity (KD),
reaction stoichiometry (n), enthalpy (ΔH), and entropy (ΔS). ITC is useful for characterizing

The HIR Nanostring (GEN2) Facility which is based on a digital molecular barcoding technology is
a multi-user facility that can deliver highly multiplexed, direct digital profiling of individual
molecules in a single reaction without amplification or bias. It application includes profiling
hundreds of gene transcripts, copy number variations, or miRNAs with high sensitivity and
precision. The NanoString nCounter gene expression system was reported to be more sensitive than
microarrays but similar to real-time PCR (Taqman and SYBRGreen).

The PROTEAN i12 IEF system is designed for isoelectric focusing (IEF) proteins in immobilized pH
gradient (IPG) strips for the first dimension of two-dimensional (2-D) electrophoresis. The PROTEAN
i12 IEF cell can run 1–12 IPG strips in 7, 11, 13, 17, 18, or 24 cm focusing trays. Each channel in the
isoelectric focusing tray is independently powered, enabling precise control over each IPG strip. This
feature makes it possible to run different sample types, different pH gradients, and multiple
protocols simultaneously.

Typhoon FLA 9500 is a variable mode laser scanner with modular access to the optical
components, providing both versatile and flexible imaging for precise quantitation of
proteins, nucleic acids, and other biomolecules.

Guidelines for using the HIR Agilent 1290 Ultra High Performace Liquid Chromatograph
(UHPLC)
The HIR Agilent 1290 UHPLC Facility is a multi-user facility which provides advanced
instrumentation to researchers for qualitative and quantitative analysis of various types of polar and
non-polar molecules of low or high molecular weight samples.
User must provide reference of journals, publication or application notes to confirm that the samples
must be amenable to LC techniques and reference to HPLC methods such as suitable mobile phase,
gradient composition, types of LC columns and detectors are available. Samples must go through
proper sample preparation or clean-up procedure prior to LC injection.
The Committee reserves the right to make any necessary changes to the guidelines from time to time
and as and when needed.

The HIR Agilent 1290 UHPLC Facility is located at Bilik Khas 3, Ground Floor, HIR building.

The Beckman Coulter Optima XPN100 Ultracentrifuge Facility is a multi-user facility which
provides instrumentation to researchers for ultracentrifugation: purify and/or separation of
viruses, subcellular organelles, macromolecules etc (SW 41 Ti, Type 70 Ti, and Type 100 Ti
rotors are available, which rated for 41 000 rpm, 70 000 rpm, and 100 000 rpm, respectively).

The Beckman Coulter Optima XPN100 Ultracentrifuge Facility is at present located at Bilik
