Product Description
TransFix® is a patented stabilisation solution that prevents cellular degradation in cerebrospinal fluid (CSF) for
analytical testing purposes.
TransFix/EDTA CSF Sample Storage Tubes consist of 5ml tubes containing 0.2ml of TransFix/EDTA.
Principle and Intended Use of TransFix/EDTA CSF Sample Storage Tubes
The active components of TransFix stabilise leukocytes and leukocytic antigens for up to 10 days. Using these
tubes to stabilise CSF leukocytes it is possible to determine cell subsets via flow cytometric analysis, up to 10
days after collection. Refer to reference for a detailed explanation of how to conduct the flow cytometric
analysis (available on www.cytomark.co.uk).
Procedure
1.
On receipt of the CSF specimen, record the volume of the sample and immediately transfer to the
TransFix/EDTA CSF Sample Storage Tube.
N.B. CSF samples should be transferred into TransFix/EDTA CSF Sample Storage Tubes
immediately after lumbar puncture, but failing this, CSF must be less than 2 hours old when it is
treated with TransFix/EDTA. Do not refrigerate the sample before treatment with
TransFix/EDTA.

Mix gently by inversion at least 10 times and store at 2-8°C for up to 10 days.
Allow the stabilised CSF sample to return to room temperature (18-25°C) before preparing it for
flow cytometric analysis.
Add 2ml of sterile filtered phosphate buffered saline (PBS), pH 7.4 to the stabilised CSF sample and
mix gently as before.
Centrifuge the sample at 540g for 5 minutes at room temperature.
Remove the supernatant and discard.
Re-suspend the cell pellet with 300µl of sterile filtered PBS, pH 7.4.
Examine the sample e.g. using flow cytometry evaluation.
N.B. The dilution factor – depending on the volume of CSF sample - must be accounted for when
cell counts from stabilised CSF samples are recorded.
N.B. Cross linking of proteins may occur when using TransFix/EDTA. We therefore recommend
that all antibody conjugates are validated in association with TransFix/EDTA prior to routine use.

Storage Conditions and Stability
TransFix/EDTA CSF Sample Storage Tubes must be stored at 2-8°C, but must not be frozen and have a shelf life
of one year from the date of manufacture or until the expiration date on the label. A certificate of analysis can
be provided with every TransFix/EDTA CSF Sample Storage Tube product.

Warning Statements
1. TransFix/EDTA CSF Sample Storage Tubes are for research use only.
2. Do not use the TransFix/EDTA CSF Sample Storage Tubes beyond the expiration date on the box
and tubes.
3. Incubation times or temperatures other than those specified may give erroneous results.
4. TransFix/EDTA is a clear green liquid. Do not use the TransFix/EDTA CSF Sample Storage Tubes if
there is a change in the physical appearance of the liquid as this may indicate deterioration and
will affect test results.
5. TransFix/EDTA treated specimens and all materials coming into contact with them should be
handled as if capable of transferring infection and disposed of with the proper precautions.
6. Avoid contact of TransFix/EDTA and TransFix/EDTA treated samples with the skin and mucous
membranes. Any contact should be washed off with cold water immediately.
7. TransFix/EDTA does not contain any antimicrobial reagents. Microbial contamination should be
avoided or erroneous results may occur.
