Summary and concept:
The diagnosis of infectious diseases has been revolutionized by the development of molecular techniques, foremost with
the applications of amplification methods. Polymerase Chain Reaction (PCR) being the forerunner. Originally PCR
evolved as a manual method which needed repeated standardization. Real time PCR a fully automated closed PCR
system now allows rapid, easy, standardized testing with several clinical applications in infectious diseases which are
approved for diagnostic use by various authorizing agencies as FDA, WHO, CE etc. The achievable high sensitivity and
ease with which the method can be used to detect any known genetic sequence have led to its wide application in the
microbiology. Real-time PCR assays have provided additional major contributions over conventional PCR, the ability to
confirm the amplification product and to quantitate the target concentration. The products of amplification generated
are usually confirmed using specific oligonucleotides or probes. However signature sequences of larger segments may
not always be accurate as amplification methods are rife with errors both internal and external. Sequencing as a
technique is the ‘gold standard’ for detecting the exact occurrence of the base pairs in the nucleotide sequences of the
segment of DNA of interest also known as the target. The classical methods of sequencing have also been very laborintensive, extremely expensive and demanding in terms of infrastructure. The Nex-Gen sequencing methods have
provided a base to allow this useful test to be applied at the clinical microbiology laboratory. Pyrosequencing is one such
Nex-Gen sequencing method which has found widespread application especially in mycobacteriology, virology,
bacteriology etc. Sequencing gives additional information up to genome level by which microbial genotyping including
resistance typing can be mapped. Most of molecular techniques start with the isolation of pure Nucleic Acid and then
downstream applications. Nucleic Acid extraction, Real Time PCR and Sequencing has major application in clinical
microbiology.

The workshop is a combination of hands-on experiments; demonstrations and panel discussion on
• Nucleic Acid isolation by manual method
• Quality assessment of Nucleic Acid
• PCR/Real time PCR, data analysis
• Pyrosequencing hands on and application in Clinical Microbiology
• Panel discussion on Relevance of Molecular tests in the ‘Real Time’ situation in a clinical Microbiology laboratory
Participants will learn about different techniques used for Nucleic Acid isolation and the basics of Real Time PCR with
emphasis on molecular workflow optimization and automation. They would get hands on experience on nucleic acid
isolation by manual method, Real Time PCR based diagnostic assay & analysis on Real Time PCR system, hands on
Pyrosequencing and its application in Clinical Microbiology This workshop on Molecular workflow solutions would help
to learn some of the aspects of basic and advanced molecular techniques used in clinical microbiology.
