SPE Introduction
Choosing an SPE Product
1. Characterize the sample. Factors such
as the analyte’s polarity relative to the
matrix, the presence of charged functional
groups, solubility, molecular weight, etc.,
determine how strongly the analyte is
retained by the packed bed.
2. Select a retention strategy. There are
two basic methods for sample treatment:
a. Select the packing bed to retain the
desired analyte. The contaminants
are washed off and the desired analyte
is then eluted for analysis.
b. Select the packing bed to retain the
contaminants and the desired analyte
passes directly through.

3. Select the proper packing type for the cleanest extract with the highest recovery.
a. Reversed-phase packings are hydrophobic, silica-based materials that retain moderately polar to nonpolar compounds from a polar matrix while washing off polar interferences. Or you can retain non-polar
contaminants while the polar compounds pass through unretained.
b. Normal-phase packings are hydrophilic, silica-based materials that retain polar compounds from a
non-polar matrix while washing off non-polar interferences. Or you can retain polar contaminants while
non-polar compounds pass through unretained.
c. Ion-exchange resins retain charged compounds or remove ionic interferences.
4. Optimize conditions for best results. Select proper bed size and suitable conditioning, wash and elution
solvents.
a. Poor sample recovery often occurs when the packed bed dimensions are not optimized. Too large a bed
results in incomplete elution while too small a bed results in incomplete retention.
b. Consider the solvent strength relative to the packing material. The final conditioning solvent should
be weak so it doesn’t act as an eluting solvent. Buffers should be used to control ionization of
potentially charged compounds.
c. Wash solvents should remove weakly retained interferences without being strong enough to elute the
analyte.
d. Elution solvents should be strong enough to completely elute an analyte in a small volume (1-2mL).

Extract-Clean™ Method
Development Kits
Reversed-Phase
Method Development Kits include a variety of packing
materials to rapidly identify the packing which provides
the cleanest extract and highest recovery for your
sample.
To isolate relatively non-polar compounds from a polar matrix
A. Conditioning
Rinse device with 3-5mL of methanol followed by
3-5mL of deionized water/buffer (do not allow bed to
dry before next step).
B. Sample Application
Apply sample to the top of the device and draw
through the packing bed. Flows of 1-5mL/min are
optimal.
C. Wash
Wash with 5mL of a polar solvent if analyte is to be
retained.
D. Elution
Elute analyte into a collection tube with 1-5mL of a
non-polar solvent.
Choose a Reversed-Phase Kit to retain moderately
polar to non-polar compounds from a polar matrix.
Choose a Normal-Phase Kit to retain polar compounds
from a non-polar matrix.
Complete Kits are available with reversed-phases,
normal-phases, and ion exchangers.
Normal-Phase
To isolate polar compounds from a non-polar matrix
A. Conditioning
Rinse device with 3-5mL of non-polar solvent.
B. Sample Application
Apply sample to the top of the device and draw
through the packing bed (1-5mL/min).
C. Wash
Wash with 5mL of a non-polar solvent if analyte is to
be retained.
D. Elution
Elute analyte into a collection tube with 1-5mL of a
polar solvent.
Ion-Exchange
To isolate charged or potentially charged compounds
A. Conditioning
Rinse device with 3-5mL of deionized water or low
ionic strength buffer (0.01M).
B. Sample Application
Apply sample to the top of the device and draw
through the packing bed at 1mL/min (ion exchange
kinetics are slower than reverse or normal-phase).
C. Wash
Wash with 5mL of deionized water or low ionic
strength buffer.
D. Elution
Elute analyte into a collection tube with 1-5mL of
buffer at high ionic strength (0.1-1M) or modified pH
(pH such that analyte is uncharged).

Drug-Clean™ phases are true copolymeric packings
with both reversed-phase and ion exchange groups.
Drug-Clean™ C is a C8/SCX cation function. This is
used to retain neutral or amine containing compounds
while allowing carboxylate groups to pass through.

Why is a “True Copolymer” Better Than a Mixed Bed?
By definition, a mixed mode packing contains two different functional groups on the same
sorbent. To minimize costs, two individual packings are sometimes blended together physically
and packed in a tube. This gives the effect of a mixed mode but in fact is a mixed bed packing
with batch-to-batch reproducibility dependent on the blending skills of the manufacturer. A
true mixed mode has the functional groups polymerized on the same silica base. This copolymer
treatment assures a more predictable packing performance which means more reproducible results
from tube to tube, batch to batch.

What is a Maxi-Clean™ Cartridge?
A Maxi-Clean™ cartridge is an alternative format for SPE. It uses the same high quality packing
materials as the Extract-Clean™ columns but has a polypropylene housing with both a female luer
inlet and a male luer outlet tip. This allows use of positive pressure from a syringe or negative
pressure from a vacuum manifold. 20µm polyethylene frits are placed at each end of the sorbent
bed. The packing material is packed and compressed to improve or optimize flow characteristics.

• Seven chemistries solve a variety of specific problems
The most difficult part of many IC applications is
eliminating interfering components from the sample
matrix. These interferences may co-elute or mask peaks
of interest, overload the column, or shorten the column
life by binding irreversibly to the column packing.
Alltech’s unique ion-exchange SPE cartridges eliminate
many of these interferences1, 2.
Each metal-free polypropylene cartridge contains
0.5mL or 1.5mL of purified polystyrene resin, contained
by 20µm polyethylene frits. The resin is functionalized
to retain specific types of components from the sample.

Alltech’s Benchtop Vacuum Station is a versatile,
powerful, user-friendly answer to most vacuum
applications. Three modes of operation offer flexibility
for different applications. “Continuous Vacuum Mode”
pulls a maximum vacuum with a 1-120 minute timer.
“Continuous Vent Mode” clears the pump heads of vapors.
“Set Point Mode” allows you to select the vacuum level,
and it maintains that vacuum level with an absolute
vacuum sensor, making runs reproducible regardless of
altitude or barometric conditions. It is the perfect pump
for filtration and solid phase extraction tasks.
This Vacuum/Pressure station is an economical choice

Product Selection
There are several basic steps to choosing the best
filtration product for your needs. This guide will help
you get started.
1. Choose a Filter Membrane
Refer to the Membrane Selection Guide and the Chemical
Compatibility Chart on the next pages to select the
appropriate membrane for your application.
Consider
a.) sample particulate size
b.) aqueous or organic nature of the sample, and
c.) the levels of solvent resistance and protein
retention you require.
Filter membranes remove particulates from a sample or
mobile phase. 0.45µm pores are commonly used to remove
sample or mobile phase particulates prior to HPLC analysis.
0.2µm pores remove bacteria from biological samples and
can be used for sterilization.

2. Choose a Membrane Diameter
Larger sample volumes require larger membrane diameters.
High particulate levels will reduce the amount of sample
filtered before a membrane becomes plugged, so use a
larger diameter membrane or a glass pre-filter to increase
filtrate volume with particulate laden samples.
3. Choose a Device
Consider the number of samples, the sample volume, and
the filtering mechanism you prefer to use.
Syringe Filters: Filtration is achieved by pushing the
sample through the membrane with a syringe or other
luer-connection device. Syringe filters allow you to
control the rate of flow, which can be critical with delicate
samples. Syringe filters also allow you to filter into nearly
any tube, vial, or column that represents the next step in
your analysis. Syringe filters with glass pre-filters are used
in the same manner as regular syringe filters. They are
especially useful for particulate laden samples.
Centrifuge Filter Tubes: Filtration is achieved by pulling
the sample through the membrane with centrifugal force.
This allows you to simultaneously filter as many samples
as your centrifuge can hold, and captures your filtrate in a
clean microtube.
Pre-Cut Membranes: Multiple diameters offer a wide range
of sample capacities, up to multiple liters. You need
a filtration apparatus, such as a filter funnel or other
vacuum-driven filtration device.
What is the advantage of a glass
pre-filter?
Pre-filters rest on top of the
filter membrane and help trap large
particulates to reduce clogging,
minimize sample loss, and extend the
membrane life. Many of Alltech’s syringe filters are
available with 1.0µm borosilicate glass pre-filters.
We highly recommend using syringe filters with
glass pre-filters for particulate-laden samples.

Why are hydrophobic polypropylene membranes better than hydrophilic membranes?
Polypropylene is a naturally hydrophobic filter membrane. Some manufacturers add a wetting
agent to make polypropylene hydrophilic. These chromatograms compare extractables using
isopropanol at 220nm from hydrophilic polypropylene membranes and Alltech’s pure hydrophobic
PolyPure II™ membranes. The large peak present from the hydrophilic polypropylene membrane is
the wetting agent, which elutes in organic solvents and can interfere with your chromatography.

Maxi-Spin™ Centrifuge Filter Tubes remove particulates
from larger sample sizes with any centrifuge that holds
50mL conical tubes. They should only be used with
a fixed-angle rotor centrifuge to prevent premature
clogging of the membrane. Maximum operating
temperature: 50°C. Maximum G-force: 2500xG.
Quickly and easily remove particulates and clarify
small sample volumes. Less than 5µL hold-up volume
provides near quantitative volumetric recovery.
Maximum G-Force: 10,000xG.

Anodisc® Membranes
Anodisc® Membranes are hydrophilic and highly
solvent resistant. They will work with both vacuum
and pressure systems, although we recommend using
a membrane support with vacuum systems. Use for

polypropylene support ring.

Vacuum Filtration
In-Line Vacuum Filtration Systems
Filter Funnels and an In-Line Filter Cap share a
common base to easily accommodate any vacuum
filtration volume.
In-Line
Filter
Cap
Filter funnel has a threaded filter base that connects
directly to any receiver with GL45 threads. It uses any
47mm diameter membrane. To use, connect the filter
base directly to your vacuum source using one of the six
tubing adapters included with each unit. Polypropylene
construction is ideal for mobile phase filtration,
degassing, and culture media filtration. Choose from 1L
and 2L funnel volumes.
Use the In-Line Filter Cap for 100mL to Multi-liter
Filtration tasks.
The In-Line system uses the threaded base of the
funnel system but replaces the funnel with the in-line
filter cap. Tubing is connected to the filter cap and
draws liquid into the system from any container. This
minimizes atmospheric exposure and handling hazards.
The polypropylene fluid path uses any standard 47mm
membrane. Alltech’s built-in priming valve restores
flow in seconds without replacing the membrane or
compromising the flow path.
