Proteomics course: gel based protein separation by twodimensional gel electrophoresis and protein characterization by MALDI-TOF/TOF mass spectrometry 15 – 18 June 2015 CIIMAR – Interdisciplinary Centre of Marine and Environmental Research, Univ. Porto Ipatimup - Institute of Molecular Pathology and Immunology of the University of Porto i3S - Instituto de Investigação e Inovação em Saúde FCUP - Faculty of Sciences, University of Porto ESTSP-IPP – School of Allied Health Technologies, Polytechnic Institute of Porto Introduction: Proteomics is a research area devoted to the analysis of proteins from complex biological samples. It has been applied to evaluate protein expression, to identify proteins and characterize their post-translational modifications (PTMs). Examples of such applications include the analysis of biochemical changes in organisms when exposed to an adverse environmental condition, allowing a better understanding on organism’s response and adaptation. Another major application of Proteomics is the discovery of novel diseaseassociated biomarkers with relevant clinical purposes. Two main approaches to undertake proteomic studies are protein separation by two-dimensional gel electrophoresis (2DE) and protein identification and characterization by MALDI-TOF/TOF mass spectrometry. Course aims: This course aims to describe the principles of 2DE and MALDI-TOF/TOF mass spectrometry as well as to provide basic training on protein separation by 2DE, peptide analysis, protein identification and characterization of post-translational modifications by mass spectrometry. English will be the speaking language. Participants: graduated students and researchers that have interests in the field of proteomics, 2DE and MALDI-TOF/TOF mass spectrometry analyses. Participants are limited to 20 but a minimum of 10 participants will be required to start the course. Equivalency to 3 ECTS can be solicited by the students following post-graduate courses at Porto University (for more information contact Alexandre Campos, [email protected]). Coordination: The course will be held in CIIMAR and Ipatimup/i3S research institutes and coordinated by Alexandre Campos (CIIMAR), Hugo Osório (Ipatimup/i3S), Marisa Freitas (Department of Environmental Health, School of Allied Health Technologies, Polytechnic Institute of Porto) and Vitor Vasconcelos (Faculty of Sciences, University of Porto and CIIMAR). Registration: the registration fee is 200 euros. To register please contact Marisa Freitas: email: [email protected] Tel: +351 223401813 Preliminary Programme: Monday, June 15th: 9.00 am – 13.00 pm: lectures (CIIMAR) 9.00 Welcome reception 9.30 Introduction to proteomics 10.10 Principles of protein sample preparation and 2DE 10.50 Coffee break 11.10 Principles of MALDI-TOF/TOF MS and protein identification 11.50 Protein quantification in proteomics 12.30 General discussion 13.00 Lunch 14.30 pm – 18.00 pm: laboratory work (CIIMAR) Sample preparation and protein separation by Isoelectric Focusing (IEF) (First dimension) Tuesday, June 16th: 8.30 am – 13.00 pm: laboratory work (Ipatimup / i3S) - Protein isolation and “in gel” digestion - Sample application in the MALDI plate - Mass spectra acquisition in MS and MS/MS modes - Mass spectra interpretation and protein identification 14.30 pm – 18.00 pm: lectures (CIIMAR) Presentations provided by guest researchers, focusing the research in proteomics and the different subjects covered by this discipline.
