DNA Sample Requirements for tGBS 1. Quality Requirements DNA Ratio of A260/A280 1.7-­‐2.0 per sample Concentration between 10-­‐100ng/ul RNase treatment to remove RNA during DNA extraction is required *** For best sequencing results we recommend using commercial kits for DNA extraction, such as Qiagen’s DNeasy kit or Qiagen’s Magattract Core kit We require high quality DNA for tGBS. This is not because the tGBS reaction per se requires high-­‐quality DNA, but because this is required to obtain accurate DNA quantifications. If quantifications are not accurate, the numbers of reads per sample will vary to an unacceptable level, reducing the quality of the tGBS analysis. If sample quality does not meet our criteria upon sample arrival, we may ask you to send tissue samples for our wetlab team to extract DNA directly at an additional cost to the customer Each “sample” should consist of DNA from a single individual (i.e., please do NOT pool individuals without prior discussion with and agreement from Data2Bio) 2. Quantity Requirements For pilot projects, we prefer to have 2 ug of genomic DNA per sample For normal projects, we would prefer to have 1 ug of genomic DNA per sample While we prefer to have 1 ug of genomic DNA per sample, If sample quantities are very limited, please contact us; we *may* be able to work with as little as 500 ng DNA per sample. 3. Shipping Please send your frozen DNA samples in Qiagen EB buffer in a carefully sealed 96-­‐well plate with dry ice via overnight express. If you are shipping from outside the US and have concerns about being able to ship samples via overnight, it would be best to ship lyophilized samples to us at room temperature to ensure sample integrity. Please see Tissue FAQ if you are sending us tissue samples to extract DNA Please send tracking numbers and an e-­‐copy of your sample list to: [email protected] (Template for sample list provided by Data2Bio) Please arrange your shipping schedule to ensure that your samples arrive on a US weekday. Please also ensure that they do NOT arrive on a US holiday.
