The ReadyPrep protein extraction kits (membrane I and II) yield fractions of varying complexity. The membrane I kit is a quick and effective protocol for isolating most membrane proteins. It does not require ultracentrifugation or preparation of density gradients. The membrane II protocol is better for isolating more complex membrane proteins. The ReadyPrep protein extraction kit (signal) is specific for isolating proteins involved in intracellular membrane trafficking and signaling pathways. Fractionation kits using differential protein solubility — The ReadyPrep sequential extraction kit and the ReadyPrep protein extraction kit (soluble/insoluble) both reduce sample complexity using differential solubilization. The sequential extraction kit enables the isolation of three different fractions of increasing solubility. These fractions are isolated sequentially, allowing the visualization of proteins that might not otherwise be seen. Increasing solubilization strength is provided through the use of stronger detergents for each subsequent fraction. In the ReadyPrep sequential extraction kit, the solution with the strongest solubilizing power is reagent 3, which contains the detergent SB 3-10. In contrast, the ReadyPrep protein extraction kit (soluble/insoluble) uses a single fractionation step and uses the detergent ASB-14, a stronger detergent than SB 3-10. The two kits can be used independently, or the rehydration/sample buffer from the soluble/insoluble kit can be used with the sequential extraction kit to create a fourth fraction for even better resolution. You have the option of purification by chromatography or preparative electrophoresis. Bio-Rad offers a complete line of chromatography instruments for high-resolution purification. Alternatively, preparative electrophoresis devices can be combined with other separation techniques to provide sample preparation schemes for numerous proteomic applications. Model 491 Prep Cell and Mini Prep Cell The Model 491 prep cell and mini prep cell use continuous-elution electrophoresis to purify macromolecules based on their relative mobility in polyacrylamide or agarose gels. Both the Model 491 prep cell and the mini prep cell allow resolution of proteins differing in molecular weight by as little as 2%. DNA fragments up to 18 kb may also be purified in a prep cell using agarose gel electrophoresis. For more information, see pages 238–239. MicroRotofor™ Cell, Mini Rotofor® Cell, and Rotofor Cell The Rotofor family of products fractionates complex protein samples that are free in solution. The nondenaturing liquid-phase IEF technique rapidly isolates proteins of interest based on pI, simplifying protein purification and maintaining biological activity. The cartridge selection includes UNOsphere™ Q, UNOsphere S, and Profinity™ IMAC media prepacked into 1 and 5 ml formats. Bio-Scale Mini cartridges are convenient, disposable, and supplied ready for use. The cartridges are designed for use with BioLogic™ systems, a peristaltic pump, or any chromatography system. Bio-Scale Mini affinity cartridges can also be run on the Profinia™ protein purification system (see pages 86–93). For simple step elution, the cartridges can be used with a Luer-Lok syringe.
