Create NeoPrep Library Prep System Sample Sheets with IEM FOR RESEARCH USE ONLY This quick reference card describes how to use the Illumina® Experiment Manager (IEM) to create NeoPrep™ Library Prep System compatible sample sheets. For more detailed general information on how to use the IEM application, see the Illumina Experiment Manager User Guide (part # 15031335). New or less experienced users are advised to read the guide before using this quick reference card. Create a NeoPrep-Compatible Sample Sheet 1 On the IEM main screen, select Create Sample Sheet. 2 On the Instrument Selection screen, select NeoPrep, and then select Next. 3 On the Workflow Parameters screen, do the following: a From the Sample Prep Kit drop-down menu, select one of the following, depending on the NeoPrep kit you are using: — TruSeq Stranded mRNA — TruSeq DNA b Type a run name, operator name, and notes. c Select the date. d Select the number of samples. The default is 16. e For TruSeq® DNA, select a 350, 550, or Mixed Insert Size. The default is 350. 4 Select Next. 5 On the Sample Selection screen: a Type a sample name for each sample. b The default well, index adapter, and index sequence are displayed for each sample. c For TruSeq DNA, if Mixed Insert Size was selected in step 3, select the Insert Size cell and select a size from the drop-down menu for each sample. 6 [Optional] Select Add Blank Row to add rows, and then repeat step 5 for additional samples. 7 Select Finish and save the sample sheet file. 8 When prompted, select Yes if you want to review the sample sheet in Microsoft Excel or No to exit the sample sheet wizard without reviewing the sample sheet. need there, contact Illumina Technical Support by email or phone.
