How to Use Odyssey® Protein
Molecular Weight Marker to Determine
Molecular Weights of Bands in Any Channel

To use a single color (700) marker to determine
the MW of bands in the 800 channel, first scan
the gel in both channels.
Acquire an image in the 700 and 800 channels.
Add the appropriate number of lanes to the
image, either one at a time or by using the
Add Multiple Lanes function.
Complete all band edits, then open the Lane
menu and select Edit Size Standards.
In the new window, select the size standard to
use. Select the lane(s) to use as the marker
lane(s) and click Snap To Lane. If the number
of bands in the marker lane(s) does not exactly
match the number of weights designated in the
Size Standard, there will be an error. For more
information, see Chapter 6: Sizing Bands in the
Odyssey 3.x Tutorial Manual.
Apply the marker, then click Apply to Both. A
warning opens:
Apply a Western Analysis to the image. Draw
the boundary and set the correct number of
lanes. Add or remove Bands by either clicking
the More/Fewer buttons, or manually. Choose
a marker set and the number of marker lanes
in the Marker Lanes group on the Western
Analysis ribbon. Drag the Marker Handle to
the appropriate lane(s) to set the marker
lane(s). See the Tutorial Guide for more

Summary
Both the Odyssey Application and Image Studio software allow the use of a marker lane
in one channel to determine the MW of bands
in another channel.
There are two advantages to using Image Studio over the Odyssey Application software:
1. Image Studio allows both channels to be
displayed as grayscale images.
2. Image Studio has the flexibility to use an
assigned marker, even if some of the bands
in the marker lane run off the bottom of the
gel.
Click OK. All sample bands in both channels
are now sized.
