Description Antifungal; yellow to orange powder; odorless or practically
odorless.
Solubility Insoluble in water, alcohol, and ether; soluble in propylene glycol;
insoluble in benzene and toluene; soluble in dimethylformamide and dimethyl
sulfoxide; slightly soluble in methanol; practically insoluble in aqueous media at
neutral pH values; 1 gram dissolves in 25 to 30mL of DMSO and 12–15mL acidic
dimethylformamide.
Stability Store 2–8ºC; decomposes gradually above 170ºC without melting; in
aqueous solution, trans­cis isomerization and epoxidation are the most likely
routes of degradation; amphotericin is solubilized by micelle formation and prone
to photlytic degradation; protect from light.
Packaging Light resistant and airtight container; refrigerate.
Therapeutic categories of use Antifungal; antiprotozoal.
Incompatibilities Follow manufacturer's directives.
Comments/Notes It is administered by IV, nasal instillation, inhalation, topically
in the treatment of superficial monilial infections and by nasal spray in the
prophylaxis of aspergillosis in immunocompromised patients. Amphotec is
Amphotericin B Cholesterol Sulfate Complex for Injection, a liposomal product
for IV adm. Upon reconstitution, Amphotec forms a colloidal dispersion of
microscopic disc­shaped particles. Label: to be administered intravenously or
topically. Used in Leishmaniasis in animals. Under no circumstances should a total
daily dose of 1.5 mg/kg be exceeded. Amphotericin B overdoses can result in
potentially fatal cardiac or cardiorespiratory arrest. Although some patients may
tolerate full intravenous doses of amphotericin B without difficulty, most will
exhibit some intolerance, often at less than the full therapeutic dose.
Amphotericin B Inhalation Powder (ABIP) has been granted orphan medicinal
product designation by the European Commission for the prevention of pulmonary
infections; in the United States, the FDA granted Fast Track designation and
Orphan Drug designation for the prevention of pulmonary fungal infections by
inhalation treatment.
