This manual is designed for the B490 and T490 series microscopes. Though both have identical bases, the B490 is equipped with a binocular head (2 eyetubes) and the T490 is equipped with a trinocular head (2 eyetubes & 1 photo/video port). Please take a few minutes to familiarize yourself with the features and functions of your new microscope. Precautions (1) As the microscope is a precision instument, always handle it with care, avoiding impact or abrupt movement during transportation. Do not shake the package. (2) Do not place the microscope in direct sunlight or in high heat. Keep it indoors in a dry and clean place with temperatures between 32°-100° F (0°-40°C), maximum relative humidity: 85% (3) Avoid touching the lenses on the objective and the eyepiece so that oil and dirt from your fingerprints do not obstruct your view. (4) Before turning the power on, make sure that the power supply voltage is consistent with the voltage of your microscope 3 1. Mechanical Stage Mechanically moves the slide along an X and Y axis for optimal positioning Nosepiece Houses the objective lenses Rotating Gemel Head Adjusts eyepieces to fit the distance between the user’s eyes for added comfort Tension Knob Adjusts the tension of the focusing knobs 7 2. Operation 490 SERIES 2-1 Assembly (1) First take the styrofoam container out of the cardboard carton and lay it on its side, paying attention to which side is labeled up. Remove tape and open the container carefully so as to avoid dropping and damaging the optical items. Check carefully to ensure that all parts and accessories are intact. (2) Check the parking list below: One Microscope Body One Binocular (B490) or Trinocular (T490) Rotating Gemel Head One Adjustable Trinocular Photo Port (for T490 only) Four Objectives: 4X, 10X, 40X, and 100X Two Pairs of Eyepieces: W10X and P16X Color Filters One Bottle of Immersion Oil One 6V/20W Halogen Spare Bulb One Spare Fuse One Dust Cover (3) Remove the microscope body from the box and remove the plastic protective covering. The body of the microscope is composed of the base, the stage, the arm, and the nosepiece. (4) Loosen the metal knob located directly above the nosepiece (the head-lock screw) completely. Place the head, flat circular side down, onto the circular opening at the top of the arm. Make sure the microscope head is securely on top of the arm and tighten the head-lock screw to lock the head in place. (5) For the T490 model only, screw the photo port onto the top of the microscope head. (6) Remove the eyetube caps and drop the desired eyepieces into the eyetubes without touching the lens. 8 2. Operation 490 SERIES (7) Screw the objectives into the microscope nosepiece from the lowest magnification to the highest, being careful not to touch the lens. (8) Plug in the microscope and turn it on. If no light emerges from the light source, adjust the dimmer knob on the side of the base. 2-2 Setting up (1) Loosen the head-lock screw and adjust the microscope head so that it is in a comfortable position for observation. Lock the head-lock screw. With both eyes open, look into the eyepieces. Adjust the interpupillary distance by holding the eyetubes and rotating the eyepieces either towards or away from one another, until one circle of light can be seen. (2) Place the specimen to be studied on a glass slide (or simply use a prepared slide), and place it on the stage, holding it sungly in place with the metal slide-holders of the mechanical stage. Using the slide controls, center the specimen over the stage opening, lining it up with the light and objective lens. (3) To adjust the illumination, slowly turn the dimmer until the desired intensity of light is achieved. 2-3 Focusing (1) Turn the nosepiece to choose an objective. It is easiest to use the lowest magnification first (usually 4X) to locate and focus the specimen. As you move up in magnification, you may have to refocus the image a little bit each time. When using the 100X objective, a little immersion oil must be put between the objective and the cover glass on the slide in order to minimize the distortion cause by air. Always wipe the oil off the objective with either a glass-cleaning paper or cloth after each use. 9 2. Operation 490 SERIES (2) Begin focusing by first looking with one eye through the eyetube without the diopter. Close your other eye. Loosen the limit-stop knob (located on the inside of the coarse-focusing knob) in order to give the mechanicalstage a full range of movement and adjust the coarsefocusing knob to bring the slide into focus. You may also need to re-center the specimen on the stage. (3) When the object is in your field of vision, adjust the fine-focusing knob until you see a clear, sharp image. BE ALERT: When moving the mechanical stage vertically, be careful not to hit the objective lens with the slide. Remember to use the limit-stop knob to avoid impact between the objective and the slide when using the 40X and 100X objectives. Lock the limit-stop knob at the position where the slide almost touches the objective. (4) Once the image is focused through the one eyepiece, look through the other eyepiece and turn the diopter until the image is clear through that eyepiece as well 2-4 Adjusting the condenser and diaphragm (1) Using the condenser-adjustment knob, you can change the distance between the light condenser and the stage. This allows you to control the concentration of the light hitting your slide. (2) By Changing the aperture of the iris diaphragm, you can adjust the background brightness. Adjust the aperture of the iris diaphragmm using the iris adjustment slider located directly under the stage. (3) If you want to use a color filter, swivel the filter holder out, towards you. You can now place the desired color filter into the circular opening. Slide the filter holder back to original position before observation. 10 2. Operation 490 SERIES 2-5 Using the trinocular port The AmScope T490 is uniquely designed so that you can focus the image through the eyepieces and the trinocular port simultaneously. This “parfocal” feature allows the images through the microscope’s eyepieces, and those displayed on your computer screen or TV to be viewed at the same time! You do not need an adapter to attach your AmScope camera to the trinocular port. However, you may need an adapter if you have a non-AmScope camera. Attaching a camera: (1) If your camera has a C-mount, simply screw the camera onto the trinocular port. (2) If your camera has a 23mm mounting size, remove the C-mount from the top of the trinocular port by loosening the screw. Then, drop the camera into the trinocular tube. (3) To focus through the trinocular port, simply turn the middle portion of the tube. 2-6 Setting the stage’s stop-limit (1) Set the stage to the maximum height desired. (2) Pull the stop-limit knob towards the stage. The stage’s stop-limit is now set, allowing you to move the stage down, but not up past this point. (3) To reset the stage’s stop-limit, simply push the stop-limit knob back towards the back panel of the microscope. The stage’s full range of motion is now restored. 11 2. Operation 490 SERIES 2-7 Adjusting focusing tension (1) To adjust the tension of the focusing knobs, first locate the black ridged tension-knob on the inside of the coarse focusing knob. (2) To decrease tension, rotate the tension adjustment forward, towards the stage. To increase tension, rotate the tension adjustment backwards, towards the back panel of the microscope. 2-8 Changing the bulb (1) Before changing the light bulb, first pull the plug out of the electrical socket and wait for the lamp to cool down in order to avoid being burned. Also, make sure to remove the eyepieces or secure them to the microscope before turning the microscope over! (2) Loosen the door-screw on the bottom of the microscope and pull open the small door. Pull the old bulb out and replace it with a new one. Close the door and tighten the screw. 2-9 Changing the fuse (1) First, remove the eyepieces or secure them to the microscope before turning the microscope over. (2) Loosen the door-screw on the bottom of the microscope and pull opUsing a flat-head screwdriver, unscrew the fuse located on the bottom of the microscope. (3)Remove the old fuse and replace with a new one. Screw the fuse back in place. 12 2. Operation 490 SERIES 2-10 Maintenance (+) All glass surfaces must always be kept clean. Fine dust on the optical surface should be blown off using a hand blower or gently wiped off with a soft lens tissue. Carefully wipe off oil or fingerprints on lens surfaces using tissue moistened with a small amount of 3:7 alcohol to ether mixture (+) Do not use this solution to wipe the surface of other components. These parts, especially those made of plastic, should be cleaned with a neutral detergent. (+) Do not assemble or disassemble the microscope’s electrical components yourself. (+) After use, cover the microscope with the provided dustcover. Make sure to keep it in a dry and clean place in order to prevent rust. 13 3.
