Introduction Team Abstract Insecticides are not only harmful to insects such as roaches but also pose a risk to mammals. We designed “Exterminator Coli”, an Escherichia coli machine to exterminate roaches while being completely safe for mammals. Roaches store their energy in the form of trehalose, which can be converted to glucose by the enzyme trehalase when needed. However, mammals store their energy in the form of glycogen, which does not require trehalase to be converted to glucose. A trehalase inhibitor can therefore be used as an insecticide that is harmless for mammals. In our project, we constructed a precursor production device and an inhibitor production Exterminator coli device that can be combined to create Exterminator Coli to produce a safe insecticide. eats meal Synthesizer for trehalose OH HO OtsA We selected 3,3‘-diketotrehalose (3,3’-dkT) as trehalase inhibitor. 3,3’-dkT shows an inhibitory effect toward trehalase because of structural similarities to trehalose (Sode et al., 2001). To produce 3,3‘-dkT in E. coli, we focused on glucose-3-dehydrogenase (G3DH), which catalyzes the oxidation of the C-3 hydroxyl group of glucosides, converting them to the corresponding 3-ketoglucosides (Kojima et al., 2001). As G3DH has remarkably wide substrate specificity, it can convert not only monosaccharides but also disaccharides, including trehalose.
