Molecular formula C17H21NO4·HBr.3H2O = 438.3
Description Colorless or white crystals or white, granular powder; odorless;
slightly efflorescent in dry air; the anhydrous salt melts about 195–199°C; pH (1
in 10 solution) 4–5.5; has a pKa = 8.01.
Solubility 1 gm in 1.5 mL water or 20 mL alcohol; slightly soluble in chloroform;
insoluble in ether.
Stability Store at controlled room temperature (between 68°F and 77°F); protect
parenteral solution from light.
Packaging Oral tablets, supplied in opaque plastic bottles of 100 Scopolamine
HBr 0.4 mg tablets; ophthalmic solution, dropper bottle, 5 ml Scopolamine HBr
0.25%, ophthalmic solution; parenteral solution for injection, supplied in 1 ml vials
containing Scopolamine HBr 0.4 mg/1mL; and transdermal patch, boxes of 10 or
24 Scopolamine 1mg/72h, patch.
History Scopolamine or hyoscine is an anticholinergic or solanaceous alkaloid in
species of the plant genuses Datura, Duboisia, Hyoscyamus, and Scopolia.
Scopolamine was available for years, prior to FDA approval in 1939. Under the
General Agreement on Tariffs and Trade (GATT), the patent for Transderm­Scop
expired in 1995. In 1997, the FDA approved scopolamine soluble tablets.
Therapeutic categories of use CNS agents, antimuscarinic
antiemetic/antivertigo agents for prevention of nausea and vomiting associated
with motion sickness and prevention of postoperative nausea and vomiting.
Incompatibilities It may precipitate at pH values > 6.0 that create >1% of the
non­ionized or free­base form, B. It may form insoluble salts with aromatic
organic anions, depending on concentration.
Comments/Notes It differs from other antimuscarinic drugs in that, in
therapeutic doses, it is a sedative and tranquilizing depressant to the CNS. In its
peripheral actions, it differs from atropine in that it is a stronger blocking agent for
the iris, ciliary body, and salivary, bronchial, and sweat glands but is weaker in its
action on the heart, in which it is incapable of exerting actions in tolerated doses,
the intestinal tract, and the bronchial musculature. It is sometimes given as a pre­
anesthetic medication for both its sedative­tranquilizing and antisecretory actions.
It is effective as a prophylactic against motion sickness, for which slow­release
transdermal dosage forms have been devised. Transdermal systems provide
sustained release for 3 days. It is also used sometimes in other types of vertigo. It
is occasionally used to suppress delirium. As a mydriatic and cycloplegic, it has a
somewhat shorter duration (3–7 days), and intraocular pressure is affected less
markedly than with atropine.
